Development and Validation of RP-HPLC Method for Simultaneous Determination of Curcumin and Quercetin in Extracts, Marketed Formulations, and Self-Nanoemulsifying Drug Delivery System

Background: Curcumin and quercetin are the two important flavonoids that being used as food ingredients across globe. They also possess many pharmacological activities such anti-inflammatory, anticancer antidiabetic activity. The combination of both these phytoconstituents is available in market. However, there lack any suitable analytical method reported for their simultaneous estimation well pharmaceutical products. Hence, a need to develop them. Objectives: study aims validate simple high-performance liquid chromatography (HPLC) determination curcumin quercetin. Method: Gradient elution was carried out ACN GAA (2% v/v) 15 min wherein ratio varied from 40-60 v/v ACN: 60-40 0-5 followed by 60-80 40-20 between 7- 10 80 % 20 10-15 min. flow rate kept 1 mL/min detection wavelength at 395 nm. has been validated according International Conference Harmonization (ICH) Q2 (R1) guidelines with respect specificity, system suitability, accuracy, precision, robustness. developed further different herbal extracts, marketed formulations containing drugs self-nano emulsifying drug delivery (SNEDDS). Results: linear concentration range 50-250 ng mL-1 800-1600 mL−1 quercetin, respectively. limit (LOD) quantification (LOQ) were 11.76 35.6 curcumin, respectively, while LOD LOQ 32.94 99.76 ng/mL−1, percentage recovery found be 95-105% relative standard deviation less than 2% indicating accuracy precision drugs. Further, specific detect presence SNEDDS. Conclusions: showed excellent linearity, precision. Thus, it can explored biological samples other formulations.
